PROTEIN KW-800 series
The packing material of the Shodex PROTEIN KW-800 series is based on silica particles whose surface is coated with a hydrophilic polymer. This column is for aqueous SEC (Gel filtration chromatography (GFC)). It enjoys a positive reputation thanks to its high recovery ratio of proteins and peptide. The PROTEIN KW-800 series comprises three kinds of columns, differing in pore and particle size.

PROTEIN KW400 series
The Shodex KW400-4F series is the high-performance semi-micro column version of the KW-800 series. The theoretical plate number and the sensitivity are improved by reducing the diameter of the packed particle to 3 micrometers. KW405-4F, the newest addition to the KW400 line-up, has a larger pore size than any columns in the existing KW-800 series, and can analyze proteins whose molecular weight is larger than 1,000,000. It enables the scientist to carry out the analysis of protein complexes.

Product instruction
Usable solvents are listed below.
(1) The standard eluent is a buffer with a salt. Buffers such as phosphate, acetate, and Tris-HCl can be used.
(2) Salts such as sodium chloride, sodium sulfate, potassium sulfate and ammonium sulfate can be used. Please keep total concentration of salts under 1 M. Their recommended concentration ranges are 0.1 - 0.3 M.
(3) Up to 100 % (v/v) acetonitrile, methanol, ethanol, and 2-propanol (IPA) are usable.
(4) Protein denaturants such as urea and 6-M guanidine hydrochloride, and surfactants such as SDS and Brij-35 can also be used. However, since protein denaturants and surfactants tend to remain on the column, solvent replacement after their use takes a longer time than replacement from general solvents. It is recommended to dedicate the column for this specific use.
<Cleaning Method>
(1) Insoluble components that block the column inlet may be removed by reversing the flow direction, i.e., introducing the eluent from the column outlet, with flow rate at less than half of the recommended flow rate.
(2) Follow below cleaning steps for adsorbing components. For an efficient cleaning, reverse the flow direction and use the flow rate at less than half of the recommended flow rate. Introduce 5 to 10 times column volume of the washing solvent.
Method 1: Adsorption of ionic (basic) compounds
· Introduce the eluent with a salt concentration of 0.5 M.
· Introduce an acidic aqueous solution of pH 3.
Method 2: Adsorption of hydrophobic compounds
· Introduce the eluent with 10 - 20 % (v/v) organic solvent (acetonitrile or methanol).
